The A4 has been closed in both directions to allow an air ambulance to land, between the junctions with the A34 and Stockcross.
The car involved was a white Mini Cooper.
Thames Valley Police and ambulance crews are in attendance.
Motorists are reporting heavy traffic in the surrounding roads as drivers try to find alternative routes ahead of the evening rush hour.
Traffic is being diverted through Stockcross and via the B4000.
Police spokeswoman Rhianne Pope said at 5.10pm that the road would remain closed until the vehicles had been recovered.
Recovery services are not yet at the scene.
