A spokeswoman for Thames Valley Police, Hannah Williams, said that the incident occurred at around 8:30am, and involved a collision between a blue Vauxhall Corsa, a white Peugeot van and a black Volkswagen Golf.
A spokesman from South Central Ambulance Service, James Keating-Wilkes, said that two people were being treated by paramedics - a woman, aged 42, with back pains and a man, aged 35, who was suffering with shock.
He added that one had been taken to Royal Berkshire Hospital.
One lane is closed, and reports suggest congestion stems back to the Tot Hill services.
